Factors affecting viability and mitotic activity of mesophyll protoplasts of Sorghum bicolor have been investigated. Protoplasts were isolated from leaves of 7-day-old seedlings which had been kept in continuous darkness for 3 days prior to enzymatic digestion. Subsequent incubation of these protoplasts in a medium conditioned by prior growth of sorghum shoot callus resulted in over 90 per cent of the original population remaining viable after 1 day and 5.2 per cent undergoing nuclear division after 17 days. Also, [('3)H]-thymidine incorporation and autoradiography confirmed that DNA synthesis was occurred in these protoplasts. Nuclear division
