Summary: This book chapter provides an overview of physical mapping in plants and its use for map-based gene cloning. A brief overview of cytogenetics-based physical mapping strategies, and physical mapping approaches currently used and the lessons learnt from the success stories were furnished. The statistical models and comparative physical mapping approaches and the future direction of physical mapping are discussed in light of the fully sequenced genomes of many major crop plants in the near future.
