Summary, etc. Stem rust (Puccinia graminis f. sp. tritici) is an important disease of both wheat and barley. Highly virulent stem rust races like TTKSK (aka isolate Ug99) are virulent on greater than 90% of the world?s wheat and barley cultivars. The objective of this research was to identify barley accessions carrying novel resistance to races in the Ug99 lineage and utilize associating mapping to identify the underlying genetic component. The USDA National Small Grains Collection (NSGC) barley core collection consisting of 2,563 accessions was evaluated and a curated subset (NSGC iCore) was used in the analysis. Accessions were screened against race TTKSK at the adult stage in Njoro, Kenya and at the seedling stage in the Biosafety Level-3 containment facility in St. Paul, Minnesota. Accessions were also screened against race QCCJB at the adult stage in St. Paul, Minnesota. The collection was split in half for testing in Njoro, Kenya: in 2012, 27% (291 accessions) had intermediate to high levels of resistance and in 2013 approximately 15% (177 accessions) displayed resistance to TTKSK at the adult plant stage. Seedling evaluations to race indicated that resistance is present in the collection with a frequency of 7% (197 accessions). At St. Paul, 6% (167 accessions) showed intermediate to high levels of resistance to race QCCJB at the adult plant stage. Association mapping detected significant loci for all traits evaluated. Loci associated with adult plant reaction to QCCJB on chromosome 5H are likely due to the rpg4/Rpg5 complex. Loci on other chromosomes for other traits were not located near genes of known function and warrant further investigation.
