Summary, etc. 【Objective】Grain hardness is an important factor in determining wheat end-use quality and one of the most important targets in wheat quality improvement. 【Method】In this study, two monocotyledonous expression vectors pUBPa and pUBPb harboring pinA and pinB genes, the major genes controlling grain hardness, were transferred into wheat variety Zhongyou 9507 (pinB-D1b) via pollen-tube pathway, respectively, and transgenic plants were obtained. 【Result】The integration of the foreign puroindolines genes was confirmed by both PCR and genomic Southern blot analysis. Quantitative analysis of the friabilin on the surface of water-washed starch granules showed that the integration of foreign puroindolines genes indeed affected the expression level of puroindolines and grain texture. 【Conclusion】In conclusion, PINA and PINB (Puroindoline) interact to form friabilin and affect wheat grain texture.
