By means of immunohistochemical alpha-amylase determination techniques and Calcofluor white M2R New staining of residual endosperm cell walls it was possible to observe transport of alpha-amylase and breakdown of cell wall material in germinating seeds of Hordeum vulgare cv. Nordal. When barley seeds of which the distal tip had been removed, were treated with gibberellic acid, the importance of the distal aleurone cells in the production of both alpha-amylase and cell wall breakdown hydrolases was clearly seen
