BYDV was purified from infected barley plants by an improved procedure whichyielded 44 æg virus/100 g tissues. The isolate had isometric particles, diam. 27.3 nm and a UV absorbance spectrum with an av. A260/A280 ratio of 1.71. In ELISA using the double antibody sandwich method, the virus reacted with homologous antiserum but not with antiserum to potato leafroll virus and vice versa. It reacted slightly with heterologous antisera to other luteoviruses suchas PLRV, soyabean dwarf and beet western yellows viruses in agar gel double diffusion tests.
